วัดศาลาปูนเป็นวัดเก่าแก่ในสมัยกรุงศรีอยุธยา ไม่ปรากฏหลักฐานผู้สร้าง เป็นที่ประดิษฐานพระพุทธรูปโบราณ นามว่าหลวงพ่อแขนลาย พระพุทธรูปหน้าตักกว้าง 29 นิ้ว พุทธลักษณะเป็นศิลปะอยุธยา คล้ายกับการสร้างพระมาลัย ภายในพระอุโบสถประดิษฐานพระพุทธรูปยืน ซึ่งถือเป็นพระประธานองค์เดียวในจังหวัดพระนครศรีอยุธยาที่มีพุทธลักษณะยืน สร้างในสมัยกรุงศรีอยุธยา
นอกจากนี้ วัดศาลาปูนยังเคยเป็นที่สถิตของสมเด็จพระราชาคณะชั้นผู้ใหญ่ ที่ได้รับพระราชทานแต่งตั้งขึ้นในเขตหัวเมืองชั้นนอก นามว่า “สมเด็จพระพุฒาจารย์ (พุก)"
